I have been an officer for Interact club for three years. This year, being president for THREE middle school branches. Next year, I have the choice of opting out or obtaining a position as web master. I've recently received two president positions for my other clubs, one being the American Red Cross. My question is if I opt out of interact, will that hurt my application? I will continue to have leadership positions regardless. Plus, I will be attending Girls State this summer..

No, it won't hurt. You are doing enough other leadership positions, plus girl's state. Remember, leadership is not really measured as cumulative (years) as much as it is a pinnacle (highest level achieved).

    drop out

It is quality vs quantity. You can devote more energy to your remaining leadership positions.
